Goodjob 操作流程
1:具備傳統(tǒng)的照膠功能,可用于化學放光分析,多色熒光,實時電泳等。
2:PCR(polymerase chain reaction):聚合酶鏈式反應。
(1)DNA變性(90-96℃):熱作用,氫鍵斷裂為單鏈DNA.
(2)退火(25-65℃):引物與DNA模板結合,形成局部雙鏈.
(3)延伸(70-75℃):Taq酶(DNA聚合酶)作用下,以dNTP為原料,從引物3-5延伸,合成與模板互補DNA.

(4) CCD power & siginal cable CCD電源&數(shù)據(jù)連接線
(5) Turn on main power、CCD、white light、mini-pc
打開主電源、CCD、白光燈、迷你電腦
(6) Auto-booting to capture window 自動啟動捕捉窗口
(7) Start to load sample active Loop icon to see realtime image 開始加載樣品,選擇Loop觀察實時圖像
Adjust: zoom,focus aperture always set to maximum.routine use don’t need to adjust,only do it when size change or out of focus.
校準:變焦,光圈通常設置為*大值。日常使用不需要校準,僅當樣品規(guī)格改變或超出聚焦范圍時校準。
(8) adjust zoom/focus 調整變焦/焦距
(9) start to capture time-lapse samples 開始延時捕捉樣品圖像
turn off loop icon 關閉Loop
click time-lapse icon 點擊Time-lapse
(10)change capture image no. 改變捕捉圖像數(shù)量
(11)set exposure time. bin no. auto-save image. Next dir 設置曝光時間、bin值、自動保存圖像、下一目錄
Pre-set 預先設置
1)auto-stretch for non-saturate bands auto-stretch用于淺色條帶
2)negative for dark bands image and white background negative 用于深色條帶和白色背景
(12)turn off reflectant white light.press start 關閉反射白光,按開始
1)the first image to last image maintain same intensity,this lead to high concentration bands not easy to saturated. **到*后一張圖像保持相同的強度,這使得高強度的條帶不易被飽和。
2)turn off auto-stretch,this lead to image more like X-ray film and easy to see high concentration bands over exposure. 關閉auto-stretch,這會使圖像更像X光片,并且很容易發(fā)現(xiàn)高強度的條帶曝光過度。
(13)choose any image after capture finish. 捕捉完后選擇任一圖像
Time-lapse capture
Expose=0.5sec./per fame
Set 0-10 images or more
Bin=1,auto-stretch.
(14) turn off white light image capturing
Time-lapse capture
Expose=1sec./per fame
Set 0-10 images or more
Bin=1,auto-stretch.
